Synthese von N-Ethyl-5-thioxo-bis[1,2]dithiolo[1,4]thiazin-3-on
(4-ethyl-5-thioxo-3H,4H,5H-bis[1,2]dithiolo[3,4-b:4',3'-e][1,4]thiazin-3-one)
N-ethyl-5-thioxo-bis[1,2]dithiolo[1,4]thiazin-3-on 1 gehört zu den Derivaten des Scorpionins 2, welches eine Bis([1,2]dithiolo)[1,4]thiazin-Grundstruktur (heterocyclisches s-Indacen-Deivat) aufweist.
Schwefelhaltige Heterocyclen wie Thiophene und 1,3-Dithiole sind aufgrund ihrer Supraleitfähigkeit ausführlich studiert und beschrieben wurden. Polyschwefel- und stickstoffhaltige Heterocyclen bilden dabei noch bessere Kandidaten, sind aber synthetisch nicht immer leicht zugänglich. 1997 veröffentlichten REES et al. ihre Arbeit zur Ein-Topf-Synthese von Bis([1,2]dithiolo)[1,4]thiazinen aus Hünig's Base (N-Ethyldiisopropylamin, DIPEA), wobei durch Reaktion des N-Ethyldiisopropylamins mit Dischwefeldichlorid und einer Base als Protonenakzeptor die Zielverbindungen in guter Ausbeute erhalten werden konnten. Als Base wurde dabei DABCO (1,4-Diazabicyclo[2.2.2]octan) eingesetzt. Später wurde gefunden, dass ein 15-facher Überschuss (da bei der Reaktion 14 Protonen gebildet werden) des N-Ethyldiisopropylamins ebenfalls zu moderaten Ausbeuten führt. Interessant ist jedoch die Wahl des Lösungsmittelds, da in nicht-sauerstoffhaltigen Lösungsmitteln, wie 1,2-Dichlorethan, das Dithioderivat und bei Verwendung sauerstoffhaltiger Lösungsmittel, wie THF, ein Thio-Oxo-Derivat gebildet wird.
Scorpionin ist nicht - wie man vermuten könnte - das Alkaloid des Skorpions sondern verdankt seinen Namen seiner Molekülstruktur, die einem Skorpion sehr ähnlich ist.
Geräte:
250 mL-Dreihalsrundkolben, Magnetheizrührer mit Ölbad, Thermometer (-40°C) Refluxanlage, Möglichkeit zur Vakuumfiltration, Septum, Messzylinder, Spritzen mit Kanülen, Kieselgur, Trockeneisbad (optional: Rotationsverdampfer)

Hinweis: Beim Arbeiten mit Trockeneis ist Vorsicht geboten. Dischwefeldichlorid sollte wie Hünig's Base unter dem Abzug gehandhabt werden.
Durchführung:
Der Rundkolben wird mit 10,4 mL N-Ethyldiisopropylamin und 100 mL trockenem THF beschickt und im Trockeneisbad auf mindestens -40 °C abgekühlt. Nachdem das Sieden des Kühlbades aufgehört hat, werden durch ein Septum über eine Kanüle 4,8 mL Dischwefeldichlorid zugetropft, wobei ein Temperaturanstieg zu verzeichnen ist. Es wird so langsam zugegeben, dass die Temperatur möglichst bei -40°C bleibt. Aus der immer gelber werdenden Lösung fällt ein Feststoff aus und die Farbe vertieft sich. Es wird noch 15 min bei -40 °C und danach für 72 h bei Raumtemperatur gerührt. Anschließend wird das Ölbad unter den Kolben gesetzt und die Mischung für 5,5 h refluxiert. Die zuvor dunkelbraune Lösung verfärbt sich während des Refluxierens grün. Gegen Ende der Reaktion setzt sich beim Ausschalten des Magnetrührers ein schwarzer, kristalliner Niederschlag ab und es verbleibt ein roter Überstand. Die Lösung wird über Kieselgel abgenutscht, wobei durch das Abkühlen des Lösungsmittels weitere (weiße) Kristalle ausfallen. Die Lösung wird erneut von diesen getrennt und das Lösungsmittel im Rotationsverdampfer entfernt, wobei ein dunkelrotes Öl verbleibt, aus dem sich partiell kleine Kristallbrocken und bei längerem Stehen große Kristallquader (bis 10 mm) bilden. In der Literatur wird das Produkt durch MPLC aufgereinigt. Da dieses Verfahren im Hobbylabor schwer möglich ist kann das Produkt auch mit Petrolether gereinigt werden. Dazu wird das rote, verbleibende Öl mit Petrolether versetzt, aufgekocht und durch Dekantieren abgetrennt. Der Petrolether wird dann in ein geeignetes Becherglas gefüllt und nach längerem Stehen kristallisiert das Produkt in Form roter Kristalle aus. Alternativ kann das rote Öl auch in Dichlormethan gegeben und durch Verdampfen und Abkühlen kristallisiert werden, wobei sich lange rote Nadeln bilden.
Ausbeute: XX,XX g (YY,Y% d.Th.)
Entsorgung:
Die Lösungsmittel werden recycelt und das Produkt aufbewahrt oder dem organischen Feststoffabfall zugeführt.
Erklärung:
Die Reaktion ist noch nicht genau geklärt, allerdings wird aufgrund einiger isolierter (grüner) Salze folgender Mechanismus angenommen. Dabei ist sehr interessant, dass in den 15 Reaktionsschritten 14 Bindungen gebrochen und 12 neue aufgebaut werden.
Wie man sieht, entscheidet die Art des Lösungsmittels erst im letzten Schritt über die entstehende Spezies. Durch die Verwendung von THF wurde hier jedoch das rote Thio-Oxo-Derivat erhalten. Das Dithio-Derivat ist hingegen schwarz und fällt (wahrscheinlich) als Nebenprodukt an und fällt in Form schwarzer Nadeln bereits beim Sieden der THF-Lösung aus. Das Dioxo-Derivat kann mit Hilfe von Ethylchloroximidoacetat und das Dithio-Derivat mit Hilfe Lawesson's Reganz aus dem Thio-Oxi-Derivat gewonnen werden.

Hi Misha: I can take a look in the Paper today, but i think with this synthesis there is no way to get the H-N-Scorpionin, 'cause for the mechanism you'll need a substituated N-Atom. Otherwise iPr2N-S-S-N-iPr2*2HCl is forming (and the reaction stops on this level.) But I think there are (unstable) H-N-Scorpionines known...

I am interesting in planar form of scorpionin and for sure N-H should be like that. So if somebody will find something about synthesis and properties I will be very thankfull. What is about first to protect N in the diisopropylamine with BOC or (Me)3SiCl (or MOM) and then run reaction. And afterwords just to cleave it?
Also is interesting properties of N- substituted scorpionin like photoluminescenc( is it emit light under UV or other illumination?). How stable it is for the air influences? Is it hygroscopic? Is it degradable during some time under air and high humidity? Do somebody has any absorbance spectrase of any scorpionin derivates?
Also interesting is how aromatic are such structures?

Here are some crystals of Et-N-scorpionine (X=S,O):
Maybe this structures (counterion maybe Tos- or Hal-) could be interesting for you, because they are planar again (or did'nt you mean only a planar 3-ring-system? xD),.. i think they should be stable, but i don't think they are fluorescent... in my opionion the Et-N-scorpionine is not fluorescent too.
